一般描述

FITC-CM-polysucroses are standard FITC-polysucrose carrying carboxymethyl-substituents. Polysucrose (renamed from Ficoll™) is a polymer which displays excellent biocompatibility. The CM (carboxymethyl) group gives the product anionic (negative) charge. All batches are checked for molecular weight, degree of substitution, loss on drying and free FITC. Aladdin produce FITC-CM-polysucroses with a molecular weight of 70 kDa and 400 kDa. FITC-CM-polysucrose is supplied as a yellow powder.

Structure

Polysucrose is a polymer synthesised by cross-linking sucrose with epichlorohydrin. FITC-CM-polysucroses are manufactured by reacting selected polysucrose fractions with an activated carboxymethyl derivative in alkali whereby O-carboxymethyl groups are introduced along the polysucrose chain. The carboxyl content is approximately 5% which is equivalent to about one CM group for every five glucose units. Thereafter, fluorescein (FITC) groups are introduced by reaction with fluorescein isothiocyanate. Polysucrose has a more spherical structure and is less flexible compared to dextran.

FITC-polysucrose has an excitation maximum of 495 and an emission maximum at 517 nm at pH 9.

Application

FITC-CM-polysucrose are mostly used for studies of permeability and microcirculation. The carboxyl groups will impart an overall negative charge to the molecule, which may be valuable in gaining information on the permeability characteristics of cell membranes and tissues. FITC-CM-polysucrose has played an interesting role in elucidating the properties of the glomerular membrane.
