ZnAF-2是一种不可透过膜的乙二氨基荧光素化合物,可作为高亲和力Zn 2+特异性荧光探针(K d = 2.7 nM)。它具有低的基础荧光,当化学计量比(1:1)与Zn 2+结合时,荧光强度增加约51倍。ZnAF-2四盐酸盐对Ca 2+,Na +,Mg 2+或K +几乎没有亲和力。ZnAF-2已成功用于监测大鼠海马样本中细胞外Zn 2+释放的时空动态。ZnAF-2的可透过细胞的二乙酰化形式也可用于细胞内。
ZnAF-2 is a membrane-impermeable ethylenediaminofluoresceinyl compound that acts as a high-affinity Zn|2+|-specific fluorescent probe (K|d|= 2.7 nM). It has a low basal fluorescence which increases in intensity ~51-fold upon stoichiometric (1:1) binding to Zn|2+|. ZnAF-2 tetrahydrochloride exhibits little affinity towards Ca|2+|, Na|+|, Mg|2+|, or K|+|. ZnAF-2 has been used successfully in monitoring spatio-temporal dynamics of extracellular Zn|2+|release in rat hippocampal samples. A cell-permeable, diacetylated form of ZnAF-2 (sc-216074) is also available for intracellular use.
